      高通量测序技术能够揭示微生物群落中物种组成及优势菌属.与传统的纯培养技术相结合,更有利于微生物的研究.结合微生物群落物种组成数据,在同一阶段筛选菌株,考察了筛选得到的菌株菌属和微生物群落中优势菌属之间的联系,同时也考察了菌株的相关特性.结果表明在同一阶段,纯培养一定程度上可以获得与微生物群落中的优势菌属对应的单菌株.Burkholderiasp.YJIP-F1的吲哚降解和靛蓝生成能力在8株菌中最优.采用响应面法(RSM)优化菌株合成靛蓝产率,最优条件为吲哚0.14g/L,苯酚0.17g/L,酵母浸粉0.16g/L.相比初始条件,靛蓝产率提高了1.43倍,为生物法合成靛蓝类色素提供了新的菌株资源.

      High throughput sequencing technology can reveal the species composition and dominant genera in the microbial community. Combining with the traditional pure culture technology, it is more conducive to the study of microorganisms. Combined with the data of microbial community composition, strains are screened at the same stage. The relationship between the screened strains and the dominant bacteria in the microbial community is investigated. The characteristics of the strains are also studied. The results show that at the same stage, pure culture could obtain strains corresponding to the dominant genera in the microbial community. Among the 8 strains,Burkholderia sp. YJIP-F1 has the best ability of indole degradation and indigo production. Response surface methodology (RSM) is used to optimize the synthesis rate of indigo. The optimum conditions are as follows: indole 0.14 g/L, phenol 0.17 g/L and yeast extract 0.16 g/L. The synthesis rate of indigo is increased by 1.43times compared with that produced under initial conditions. The results provide a new strain resource for the biosynthesis of indigo pigments.
